DNA gel electrophoresis is a standard tool of biochemistry and molecular biology laboratories. The common dye ethidium bromide suffers from toxicity concerns and requires the use of damaging ultraviolet light. We observe that exposing plasmid DNA to a UV transilluminator for only 1 s results in detectable loss of colonies following transformation, suggesting rapid accumulation of DNA damage. SYBR Safe, a commercial product, is marketed as a safe alternative to ethidium bromide and has excellent sensitivity with nondamaging blue light, but suffers from prohibitively high costs. We show that thiazole orange, the parent compound of SYBR Safe, is an excellent, simple, and inexpensive alternative to these dyes. It is excitable with safe blue light or UV light, with DNA detection limits in agarose gels similar to ethidium bromide and SYBR Safe (1–2 ng/lane). Thiazole orange safely allows the use of nondamaging blue light at the same cost as ethidium bromide.  相似文献

以钛酸四丁酯为前驱体,通过溶胶-凝胶法制备了碳纳米管(CNT)改性的TiO2光催化剂.考查了不同CNT含量的光催化剂的催化活性,结果农明CNT的质量分数为0.3%时对甲基橙具有最佳的降解效果,100min的降解率接近90%,明显高于纯TiO2.采用XRD、IR和UV—vis对光催化剂进行了表征,光催化剂的粒径经计算为20nm~30nm,红外光漕中改性TiO2有羰基吸收峰,紫外光谱中改性TiO2的吸收出现明显红侈.  相似文献

Monodisperse silica spheres that comprised a rhombic‐dodecahedral zeolitic imidazolate framework core–shell microsphere were applied in the sample pretreatment of navel orange. A rapid and efficient liquid chromatography with triple quadrupole tandem mass spectrometry method was established for simultaneously quantifying four plant growth regulators, 6‐benzylaminopurin, indole‐3‐acetic acid, indolepropionic acid, 3‐indolebutyric acid, in navel oranges. A satisfactory result was obtained, i.e., the peak area of the four plant growth regulators against concentration was linear with good correlation coefficients of 0.99987–0.99991. Under optimized conditions, the limits of detection were 3.0–59.4 μg/L for the four plant growth regulators. This method was applied to the simultaneous analysis of the four plant growth regulators in commercial samples, and all the detections were confirmed by acquiring transitions for each pesticide in the samples.  相似文献

The objective of this research was to use combustion synthesis to create a nano‐sized ZnO photocatalyst using citric acid as the fuel and zinc nitrate as the oxidant. The starting materials were mixed in a stoichiometric ratio, and a slurry precursor with high homogeneity was formed. The precursor was ignited at room temperature, resulting in dry, loose, and voluminous ZnO powders. The powders, characterized by SEM, TEM and XRD, showed a particle size range of 40 to 80 nm with a wurtzite structure. The ZnO powders were introduced as a photocatalyst for the degradation of methyl orange, which was adopted as a model compound. UV light (6W) was used as the irradiation source to induce synthesized ZnO powders to perform catalytic activity. The photocatalytic reaction was executed in 40 mL of a 10 ppm methyl orange aqueous solution under 254 nm UV illumination. In this work, it was observed that both UV light and ZnO powders are needed for the photocatalytic reaction. In addition, it was found that increasing the amount of ZnO powder present in the MO (methyl orange‐C14H14N3NaO3S) solution did not correlate directly with an increase in photocatalytic ability. It was found that the scattering problem of UV light also needs to be considered. The optimized photocatalytic degradation ratio in this work reached 92.7%.  相似文献

We prepared reversible micelles with dye loaded on the shells or cores by ion exchange. A poly(4-styrylmethyl triphenylphosphonium chloride)-block-polystyrene diblock copolymer prepared from poly(4-vinylbenzylchloride)-block-polystyrene and triphenylphosphine was reacted with methyl orange in a mixed solvent of benzene and acetonitrile to produce poly[4-styrylmethyl triphenylphosphonium 4-(4-dimethylamino)phenylazobenzenesulfonate]-block-polystyrene. The loading of the methyl orange on the micellar shells or cores was dependent on the composition of the mixed solvents. Dynamic light scattering demonstrated that the loading of the dye on the cores significantly expanded the micelles when compared to that on the shells. The loading of the dye on the cores shifted the UV wavelength of the methyl orange, whereas that on the shells produced no changes in the UV wavelength. Transmission electron microscopy confirmed the formation of the spherical micelles.  相似文献

A new chemosensor for Cu2+ was synthesized based on 1,2,3,4,5,6,7,8,9,10‐decahydroacridine‐1,8‐dione dyes, which exhibited an obvious fluorescent selectivity to the sensing of Cu2+ ions over other cations, such as Na+, K+, Ca2+, Cd2+, Co2+, Hg2+, Mg2+, Mn2+, Ni2+, Zn2+, Ag+ and Pb2+. Moreover, it presented a fluorescent switch function when EDTA was added to the compound‐Cu2+ complex in examined systems.  相似文献

以吖啶橙(AO)作为光谱探针, 采用UV和荧光光谱等方法研究了亚甲基蓝(MB)与鲱鱼精DNA的作用机制. 确定了在低浓度MB时, MB与DNA以嵌插方式作用; 而在高浓度MB时, MB与DNA之间为混合作用方式. 结合比n(MB)∶n(DNA)=10∶1, 结合常数 =2.46×105 L•mol-1, MB-DNA复合物的表观摩尔吸光系数ε=5.70×106 L•mol-1•cm-1. 同时研究了酸度和温度等对MB与DNA相互作用的影响, 热力学研究推导了MB结合DNA为焓驱动反应.  相似文献

A new donor moiety, 7,7,13,13‐tetramethyl‐7,13‐dihydro‐5H‐indeno[1,2‐b]acridine (IAc), was developed to control the highest occupied molecular orbital (HOMO) dispersion of thermally activated delayed fluorescent (TADF) emitters. The IAc unit expanded the HOMO dispersion of the emitters and increased the quantum efficiency of the TADF devices up to 20.9 %.  相似文献

采用固相法合成AgNbO3/石墨烯复合纳米材料,利用透射电子显微镜(TEM)及紫外-可见漫反射光谱(UV-Vis)对样品的形貌及光学性质进行了表征。研究发现,AgNbO3与石墨烯复合后,带隙能明显降低,吸收光波长范围增大。以甲基橙溶液的降解为光催化模型反应评价了AgNbO3/石墨烯复合纳米材料的可见光催化性能。结果表明:与纯AgNbO3相比,AgNbO3/石墨烯复合纳米材料对甲基橙的可见光催化性能明显增强。实验条件下,经300 ℃煅烧的AgNbO3/石墨烯(2:1)复合纳米材料表现出最优的催化性能,它对甲基橙的可见光催化脱色速率系数约为纯AgNbO3的10倍。光催化降解机理研究表明,促使甲基橙降解脱色的主要活性物种为·O2- 和 h+。  相似文献
